It is also frequently incorporated into maintenance regimens to prevent relapse after initial clearance of acne with other therapies.

Dosage and direction

For topical use only. Not for ophthalmic, oral, or intravaginal use.

  1. Wash the affected area with a gentle, non-medicated cleanser and pat dry.
  2. Apply a thin, pea-sized amount of the product to the entire face, avoiding the eyes, lips, and mucous membranes. For larger areas like the chest or back, use an amount proportional to the size of the area.
  3. Spread evenly, using just enough to cover the treatment area with a light film. Excessive application will not increase efficacy but may significantly increase the risk of irritation.
  4. Use once daily, in the evening, as part of a regular skincare routine.
  5. Consistency is paramount. Therapeutic results are typically observed after 8 to 12 weeks of continuous use.

Precautions

  • This medication may cause initial skin irritation, including redness, scaling, dryness, stinging, or burning. These effects often diminish with continued use.
  • Minimize exposure to sunlight and UV rays (e.g., tanning beds). Adapalene can increase photosensitivity. Use a broad-spectrum sunscreen (SPF 30 or higher) and wear protective clothing daily.
  • Avoid application on sunburned, windburned, dry, chapped, irritated, or broken skin, or on open wounds.
  • Extreme weather conditions, such as wind or cold, may be more irritating to skin treated with adapalene.
  • Avoid contact with the eyes, eyelids, and mouth. If contact occurs, rinse thoroughly with water.
  • Do not use waxing as a hair removal method on treated areas due to an increased risk of skin abrasion and peeling.

Contraindications

Differin is contraindicated in individuals with a known hypersensitivity to adapalene or any other component of the formulation. Its use should be avoided during pregnancy unless the potential benefit justifies the potential risk to the fetus, as retinoids may cause harm. While systemic absorption of topical adapalene is minimal, caution is advised. It is also not recommended for use by individuals who are breastfeeding without consulting a healthcare provider.

Possible side effect

The most common side effects are local cutaneous reactions, which are often dose-dependent and may peak within the first 2-4 weeks of therapy.

  • Very Common (>10%): Dry skin, skin scaling, skin erythema (redness), and a burning/stinging sensation at the application site.

Drug interaction

Formal drug interaction studies have not been conducted with topical adapalene. However, based on its properties, the following potential interactions should be considered:

  • Other Topical Products: Concomitant use of other potentially irritating topical products (e.g., medicated or abrasive soaps, cosmetics with strong drying agents, products with high concentrations of alcohol, astringents, spices, or lime) may increase skin irritation. Use with caution.
  • Other Topical Acne Medications: Using other topical acne medications, particularly those containing sulfur, resorcinol, or salicylic acid, may lead to cumulative irritation. Benzoyl peroxide can be used concurrently but may oxidize adapalene if applied simultaneously; applying one in the morning and the other in the evening is often recommended.
  • Photosensitizing Agents: Concomitant therapy with other agents that increase photosensitivity (e.g., thiazides, tetracyclines, fluoroquinolones, phenothiazines, sulfonamides) could potentiate the risk of phototoxicity.

Missed dose

If a dose is missed, apply it as soon as remembered. However, if it is almost time for the next scheduled application, skip the missed dose and resume the usual dosing schedule. Do not apply a double dose to compensate for a missed one.

Overdose

Topical application of adapalene is unlikely to produce systemic effects due to low systemic absorption. However, excessive topical application may lead to severe erythema, scaling, and skin discomfort. In case of accidental ingestion, seek medical attention immediately. Gastrointestinal irritation, including nausea and vomiting, could occur.

Storage

  • Store at room temperature between 20°C to 25°C (68°F to 77°F). Excursions are permitted between 15°C and 30°C (59°F and 86°F).
