Tastylia Oral Strip: Fast-Acting Tadalafil for Erectile Dysfunction
Tastylia Oral Strip represents a significant advancement in the delivery of tadalafil for the treatment of erectile dysfunction. This innovative oral thin film formulation is designed to dissolve rapidly on the tongue without the need for water, offering a discreet and convenient alternative to traditional tablets. It contains the same active pharmaceutical ingredient found in conventional tadalafil medications, ensuring proven efficacy in facilitating blood flow to the penile tissues. This format is particularly beneficial for patients who experience difficulty swallowing pills or seek a more rapid onset of action, making it a versatile option within a comprehensive treatment plan.
Features
- Active Ingredient: Tadalafil 20 mg per strip.
- Formulation: Oral dispersible film.
- Administration: Buccal; placed on the tongue to dissolve.
- Flavor: Typically mint or a neutral flavor to enhance patient compliance.
- Excipients: May include film-forming polymers, sweeteners, saliva-stimulating agents, and stabilizing compounds.
- Packaging: Individually sealed blisters to protect stability and ensure hygiene.
Benefits
- Rapid onset of action due to buccal absorption, potentially bypassing first-pass metabolism.
- High degree of discretion and portability, allowing for administration without water.
- Improved convenience and compliance for patients with dysphagia (difficulty swallowing).
- Predictable and long-lasting efficacy for up to 36 hours, supporting spontaneous sexual activity.
- Avoids the gastrointestinal variability sometimes associated with traditional oral tablets.
Common use
Tastylia Oral Strip is indicated for the treatment of erectile dysfunction (ED) in adult men. Erectile dysfunction is the consistent or recurrent inability to achieve and/or maintain a penile erection sufficient for satisfactory sexual performance. The medication works by inhibiting the phosphodiesterase type 5 (PDE5) enzyme, which promotes relaxation of the smooth muscles in the corpus cavernosum of the penis and increases blood flow, thereby facilitating an erection in response to sexual stimulation.
Dosage and direction
The recommended dose is one 20 mg strip, taken prior to anticipated sexual activity. The strip should be placed on the tongue and allowed to dissolve completely; swallowing is not required. It may be taken with or without food. The dosing should not exceed one strip in a 24-hour period. A dose of 10 mg may be considered for patients in whom 20 mg is not tolerated. The medication requires sexual stimulation to be effective. Healthcare providers may recommend daily use at a lower dose for some patients; this regimen must be strictly supervised by a physician.
Precautions
- Tastylia can potentiate the hypotensive effects of nitrates and is absolutely contraindicated in patients using any form of organic nitrates.
- Use with caution in patients with underlying cardiovascular disease, as sexual activity poses a potential cardiac risk. A cardiovascular status assessment is recommended prior to initiating any treatment for ED.
- Not recommended for use in patients with severe hepatic impairment or severe renal impairment (CrCl < 30 mL/min).
- Patients with anatomical deformation of the penis (e.g., angulation, cavernosal fibrosis, or Peyronie’s disease) or conditions predisposing them to priapism (e.g., sickle cell anemia, multiple myeloma, or leukemia) should use this medication with caution.
- As with other PDE5 inhibitors, Tastylia has vasodilatory properties, which may result in transient mild decreases in blood pressure.
Contraindications
- Concomitant administration of any form of organic nitrate or nitric oxide donors (e.g., amyl nitrite).
- Known serious hypersensitivity to tadalafil or any of the excipients in the oral strip.
- Patients with a history of non-arteritic anterior ischemic optic neuropathy (NAION), which has been reported rarely in association with PDE5 inhibitor use.
Possible side effect
While generally well-tolerated, the following side effects have been associated with tadalafil:
- Very Common (≥1/10): Headache, dyspepsia.
- Common (≥1/100 to <1/10): Back pain, myalgia, nasal congestion, flushing, pain in extremity, dizziness.
- Uncommon (≥1/1,000 to <1/100): Gastroesophageal reflux disease, visual disturbances (e.g., blurred vision, cyanopsia), palpitations, tachycardia, hypotension.
- Rare: Priapism (a prolonged and painful erection lasting more than 4 hours; requires immediate medical attention to prevent permanent tissue damage), hearing loss, NAION.
Drug interaction
- Nitrates: Concomitant use is contraindicated due to the risk of life-threatening hypotension.
- Alpha-blockers: Co-administration can lead to symptomatic hypotension. Dose adjustment or separation of dosing may be necessary.
- Potent CYP3A4 Inhibitors (e.g., ketoconazole, ritonavir, clarithromycin): May increase tadalafil exposure. A dose reduction of Tastylia is recommended.
- Antihypertensives: Tadalafil may add to the blood-pressure-lowering effect.
- Alcohol: Excessive consumption can increase the risk of orthostatic hypotension and dizziness.
Missed dose
Tastylia Oral Strip is not intended for chronic daily dosing in an on-demand regimen. If a dose is missed, it is not to be doubled. The next dose should be taken as needed prior to the next anticipated sexual activity, adhering to the minimum 24-hour interval between doses.
Overdose
In cases of suspected overdose, standard supportive measures should be adopted. Given that tadalafil is highly protein-bound, it is not expected to be dialyzable. The most likely manifestations of overdose would be an increase in the reported dose-dependent adverse effects, such as severe headache, dyspepsia, back pain, and profound hypotension. Medical attention should be sought immediately, with a focus on managing hypotension through fluid administration and, if necessary, vasopressor therapy.
Storage
- Store in the original blister packaging at room temperature (15°C to 30°C).
- Protect from moisture and light.
- Keep out of reach of children and pets.
- Do not use if the blister seal is broken or the strip appears damaged.
